BSS – Bee Semen Solution

BSS (Bee Semen Solution) is a sterile, colored, and hyperosmotic formula specifically designed for instrumental insemination of queen bees.

It is used to homogeneously mix semen from multiple drones, facilitating controlled polyandrous insemination that mimics the genetic diversity of a natural mating flight. The colored solution allows for easy visual mixing without vigorous agitation.

BSS acts as a buffer solution, maintaining the ideal pH and osmotic pressure to preserve sperm viability during semen collection, dilution, transfer, and storage.

It is also recommended to moisten all surfaces that come into contact with semen (e.g., syringes, capillaries, tools) with BSS beforehand to prevent cellular damage due to adhesion or osmotic shock.

The mixture should be swirled gently (avoiding bubbles) and can be reconcentrated via low-speed centrifugation. Literature advises not exceeding 1,000 xg, as higher speeds can damage up to 50% of viable sperm. A variable-speed centrifuge is highly recommended.

For storing washed semen in BSS, keep it at 14–17 °C in sterile conditions. Unopened BSS remains effective for up to 24 months (4–25 °C). Once opened or mixed with antibiotics, it should be used the same day—even though the label allows up to 2 days, sterility is no longer guaranteed after 24 hours.
